چکیده
Acknowledgements At the outset, I would like to express my deep appreciation and gratitude to Prof. Dr. Hartmut Wekerle for offering me a graduate fellowship that enabled me to work in his department. I would like to thank him for his critical comments and continuous support. While instilling a tough scientific attitude in me, he was also able to maintain a relaxed atmosphere in the lab, which made it not only stimulating to work there but also fun. I am also grateful to Dr. Andreas Holz who supervised this thesis. As my supervisor, he constantly supported, encouraged, and taught me how to focus my research. His constructive comments and constant encouragement brought out the best in me. Our hour-long discussions have sharpened my thinking about scientific problems. My special thanks to Prof. Dr. Hans Lassmann for the excellent histological analysis. I would like to thank Bernadette Pöllinger for her helpfulness and patient open ear for my problems. The list of my indebtedness to her is endless, especially for her help in correcting my thesis. My sincere thanks to Irene Arnold-Ammer for her excellent technical assistance. She also helped to maintain the relaxed atmosphere in the lab with her sense of humor. I am grateful to Prof. Dr. Elisabeth Weiß for agreeing to be an examiner of my thesis. I wish to extend my thanks to PD Dr. evaluating my thesis. I thank Dr. Anjali (IISc, Bangalore) and my dear friends Raghu, Madhavi, and Rajesh for helping me to come to Germany. My life would have been very difficult without my friends Sham, Sona, to mention only a few. Special thanks go to my wife Jeeva for her love and care.

Spontaneous opticospinal encephalomyelitis in a double-transgenic mouse model of autoimmune T cell/B cell cooperation.
We describe a double-transgenic mouse strain (opticospinal EAE [OSE] mouse) that spontaneously develops an EAE-like neurological syndrome closely resembling a human variant of multiple sclerosis, Devic disease (also called neuromyelitis optica). Like in Devic disease, the inflammatory, demyelinating lesions were located in the optic nerve and spinal cord, sparing brain and cerebellum, and the m...
متن کاملA mighty mouse: building a better model of multiple sclerosis.
The 2 cardinal cell populations mediating adaptive immunity are T and B lymphocytes. These cells play important but poorly understood roles in the immunopathological demyelinating disease multiple sclerosis (MS) and in a widely used animal model of human MS known as EAE. In the current issue of the JCI, 2 research teams report their parallel studies of double-transgenic mice expressing T and B ...
متن کاملSpontaneous relapsing-remitting EAE in the SJL/J mouse: MOG-reactive transgenic T cells recruit endogenous MOG-specific B cells
We describe new T cell receptor (TCR) transgenic mice (relapsing-remitting [RR] mice) carrying a TCR specific for myelin oligodendrocyte glycoprotein (MOG) peptide 92-106 in the context of I-A(s). Backcrossed to the SJL/J background, most RR mice spontaneously develop RR experimental autoimmune encephalomyelitis (EAE) with episodes often altering between different central nervous system tissues...
متن کاملO 3:Therapeutic Potential of a Novel NMDA Receptor Subunit 2B Antagonist in a Mouse Model of Autoimmune Neuroinflammation
Glutamate-mediated excitotoxicity and neurodegeneration have been shown as pathophysiological hallmarks of multiple sclerosis (MS) and other autoimmune inflammatory CNS disorders. N‑Methyl‑D‑Aspartate (NMDA) receptors play a pivotal role in the mediation of neuronal glutamate excitotoxicity leading to cellular damage and apoptotic cell death. Current treatment approaches targeting glutamate exc...
متن کاملP-111: An Attempt to Facilitate the Production of Transgenic Mouse As A Model for Gene Therapy of Gaucher Disease
Background: Gaucher disease is an autosomal recessive inherited lysosomal storage disorder that affects many of the body's organs and tissues by defective function of the catabolic enzyme β-glucocerebrosidase. Gene therapy is one of the efficient ways for treatment of this disease. Due to the lack of appropriate animal models, in the field of gene therapy little progress has been done.Mate...
